It seems possible the movie’s setting is a nod to the earlier game.
However, there’s a noticeable difference between the colosseum in Mario Galaxy and the “Galactic Colosseum” level from Mario Kart Wii. The Mario Kart Wii version is a flat, rainbow-colored arena with a space background.
The Colosseum shown in the Super Mario Galaxy poster is designed with a strong Roman style. It resembles a classic, round arena, featuring many arched entrances around its outer walls.
The Wario Colosseum Theory

The fact that the colosseum shown on The Super Mario Galaxy Movie poster isn’t exactly like the “Galactic Colosseum” from Mario Kart Wii doesn’t mean Mario Kart won’t still be part of the new film.
The setting might combine elements from the battle map in Mario Kart Wii and the “Wario Colosseum” track from Mario Kart: Double Dash. The “Wario Colosseum” level strongly resembles a classic colosseum, featuring the circular building and arched entrances seen on the movie’s promotional poster.
The heart of Wario Colosseum is a winding racetrack built from metal grates. A large sphere stands prominently in the center, serving as the arena’s main attraction.
The mention of “Wario Colosseum” suggests Wario, the popular Nintendo character, might have a significant role in the upcoming movie when it releases in April, even though his appearance hasn’t been officially confirmed.
